    The study aimed to partial purify of Leucine aminopeptidase (LAP) from sera of patients with hyperthyroidism and hypothyroidism.In this study, specimens were collected during the period from 1st Nov 2017 until the end of Feb 2018 at Endocrine and Diabetes Center \/ Baghdad.\u00a0 Samples of blood were collected from fifty healthy control and two hundred patients, patients divided into four groups consisted of (50) with hyperthyroidism, (50) with hypothyroidism, (50) with sub clinical hyperthyroidism and (50) with sub clinical hypothyroidism. The aged for all subjects ranged between (15-60) years with body mass index ranged (25- 31) kg\/m2.<\/span><\/h3>\n

    \u00a0\u00a0\u00a0\u00a0 The study was included partial purified of LAP from sera of (hyperthyroidism and hypothyroidism) patients with some techniques which included precipitation with ammonium sulfide (35% – 75%),ion exchange chromatography using DEAE- Sepharose column that detected four protein peaks only two peaks have LAP activity, the fractions containing LAP activity from each peak was applied to sepharose 6B column and only one protein peak that have LAP activity was obtained from each peak, with purification fold (14.13 , 19.88), yield (29.72, 31.25) for hyperthyroidism and hypothyroidism respectively.<\/span><\/h3>\n

    \u00a0\u00a0\u00a0\u00a0\u00a0 The SDS-polyacrylamide gel electrophoresis presented only one protein band for each peak with a molecular weight (~ 130KD, ~ 63KD) of LAP that purified from sera of hyperthyroidism and (~ 125KD, ~ 61KD) with the single band to LAP which it purified from sera of hypothyroidism, these peaks could be dimer and monomer of LAP.<\/span><\/h3>\n

    Also, the study was included evaluation of some biochemical parameters in the sera of (sub clinical hyperthyroidism, hyperthyroidism, sub clinical hypothyroidism, hypothyroidism) patients which included estimation the level of (TSH, FT3, FT4, LAP) and some antioxidants (Malondialdehyde, Peroxynitrite) and compared with the healthy control<\/span><\/h3>\n

    The results of Sub clinical hyperthyroidism patients were showed a highly significant increase (p\u22640.001) in the level of LAP, peroxynitrite and MDA levels in the patient\u00b4s group compared with the control group, while a highly significant decrease in TSH levels was found in the patient group compared with the control group.<\/span><\/h3>\n

    There was a highly significant elevation(p\u22640.001)in each of FT3, FT4,LAP,peroxynitrite and MDA levels in the hyperthyroidism patients\u00a0 compared with the control group, while a significant decrease(p\u22640.05) was noticed in BMI and a highly significant decrease(p\u22640.001) in TSH levels, in the patient\u00b4s group compared with the control group, also the results of sub clinical hypothyroidism patients showed a highly significant elevation (p\u22640.001) in TSH, LAP, peroxynitrite and MDA levels in the patient\u00b4s group compared with the control group<\/span><\/h3>\n

    \u00a0In hypothyroidism patients result showed a highly significant decrease (p\u22640.001) in FT3,FT4 levels in the patient group compared to the control group, while a significant elevation in BMI and a highly significant elevation (p\u22640.001) in TSH, LAP, peroxynitritr and MDA levels was found in the patient group compared with the control group.<\/span><\/h3>\n

    The correlation coefficient (r) between the LAP level and other parameters calculated with regression plot showed in sub clinical hyperthyroidism group asignificant positive correlation between LAP with (FT3, FT4) in the control and a negative correlation in patient group, also, the results showed negative correlation of LAP with TSH in the control and patients groups.<\/span><\/h3>\n

    \u00a0In hyperthyroidism was a significant positive correlation of LAP with FT3 in the control group and patients group with a significant positive correlation of LAP with FT4 in the control group but negative correlation in the patient group, also, results showed a significant negative correlation of LAP with TSH in the control group and positive correlation inpatient group<\/span><\/h3>\n

    In sub clinical hypothyroidism was a significant positive correlation of LAP with FT3,FT4 in the control and patient groups and a significant negative correlation of LAP with TSH in the control and patient groups, but in hypothyroidism results showed a significant positive correlation of LAP with FT3in the control group and a significant negative correlation in the patients group, also,there was a significant positive correlation of LAP with FT4 in the control and patient groups as well,a significant negative correlation of LAP with TSH in the control group and a significant positive correlation in the\u00a0 patient group was showed.<\/span><\/h3>\n

    This is the first study of aminopeptidase (LAP) purification from thyroid patients. It is possible that the enzyme has been purified in both dimer and monomer form.
