    This is the first study in Iraq and the world the study was designed to showed the effect of delivery nanoparticle of amygdalin extract from local apricot kernel as metabolic therapy for cancer disease<\/span><\/h3>\n

    Part one:\u00a0\u00a0\u00a0\u00a0 Fasting venous blood samples were taken from 80 subjects Baghdad teaching hospital that collected\u00a0 from Iraqi\u00a0 patients at suffering \u00a0from\u00a0\u00a0\u00a0 breast Cancer \u00a0,and controls, They have been classified into two groups as follows:-<\/span><\/h3>\n

    1. \n

      A) Patients of breast group: includes (50) female patients, with age range (35-65) years.<\/span><\/h3>\n<\/li>\n

      B) Control group: – includes (30) healthy individuals from female sex, with age range)30-45) years who have no previous diseases which may interfere with the parameters that analyzed in this study.<\/span><\/h3>\n<\/li>\n<\/ol>\n

      \u00a0\u00a0\u00a0\u00a0\u00a0 All blood samples were experiment a analyzed for amygdalin compound and amygdalin as silver nanoparticle and determination activation this compounds on Lactate dehydrogenase\u00a0(LDH\u00a0) , alkaline phosphates (ALP) , aspartate aminotransferase (AST), alanine aminotransferase (ALT), carcinoma 15.3 (CA15.3), and carcinoma 125 (CA125),<\/span><\/h3>\n

      \u00a0Results of this study showed be 600.2\u00b162.0 U\/L, 396\u00b174 U\/L, 85\u00b110.1 U\/L, 95.4\u00b113.4U\/L, 85.1\u00b110.1U\/L and 52\u00b19.2U\/L before the start of treatment courses. The level of serum with \u00a0\u00a0LDH, ALP, AST, ALT, CA15.3, CA125 level was found to be more than that in the\u00a0 \u00a0normal reference range .upon treatments with amygdalin was found no change was detected \u00a0in concentration of\u00a0 LDH, ALP, AST, ALT, CA125 however \u00a0decrease in the concentration of \u00a0CA15.3 was found to indicated 3.4\u00b10.4 U\/L while treatments with amygdalin as silver nanoparticles was found no change in the concentration of\u00a0 ALP, AST, ALT, CA125 but\u00a0 LDH and\u00a0 CA15.3 was decreased to 170\u00b122.7 U\/L and 0.9\u00b10.23U\/L respectively\u00a0<\/span><\/h3>\n

      Part two:<\/span><\/h3>\n

      The cytotoxic activity of amygdalin and amygdalin as silver nanoparticle on A594, PC3, A2780 and MCF-7 tumor cell lines was investigated in vitro using MTT assy. The amygdalin as silver nanoparticle showed the highest anti-tumor activity and indicted reduction in cell viability against all cancer cell line in a dose-dependent pattern compared with the control WRL-68. The Amygdalin significantly exhibit the most potent cytotoxic activity onA594,PC3, A2780 and MCF-7 tumor cell lines \u00a0with IC50 value of 185.9, 195.8,190.4,131.2 \u00b5g\/ml respectively, whereas IC50 values of controls \u00a0were 224.1,\u00a0 205.4 , 224.1, 205.4 \u00b5g\/ml, respectively. The Amygdalin as silver nanoparticles, significantly exhibit the most potent cytotoxic activity onA594,PC3, A2780 and MCF-7 tumor cell lines\u00a0 with IC50 value of 166.1, 186.1, 185.9 , 84.4, respectively whereas IC50 values of controls\u00a0 were 205.1<\/span><\/h3>\n

      The effect of amygdalin as silver nanoparticleon MCF-7 cell line (regarding cell count viability, nuclear intensity, cell membrane permeability, mitochondrial membrane potential and cytochrome c) were further verified using High Content Screening array scan with multi-parametric kit. Results indicated that the amygdalin as silver nanoparticledose-dependently inhibited the cell growth of MCF-7 cells with a maximum inhibition of 54.5% at 400\u00b5g\/ml. The intensity of cell membrane permeability was also increased in a dose-dependent pattern by increasing the concentration amygdalin as silver nanoparticles ,<\/span><\/h3>\n

      On the other hand, only at higher concentration (400\u00b5g\/ml) amygdalin as silver nanoparticlescaused the reduction in mitochondrial membranepotential compared with positive control of 20\u00b5M doxorubicin by 56.6%\u00a0 and\u00a0 73.7.6% , respectively n which the disruption of mitochondrial membrane potential in consequence resulted in the release of the mitochondrial cytochrome C, which was significantly detected by 2.1 fold increases in the average fluorescent intensity of the treated MCF-7 cells than that of untreated cells<\/span><\/h3>\n","protected":false},"excerpt":{"rendered":"
