the study was included, 56 samples, 37 from patients with non-Hodgkin’s lymphoma (NHL) and 19 from normal lymphoid tissue that were used as control. The samples were collected from the educational laboratories and the Martyr Ghazi Al-Hariri Hospital in Baghdad Medical City and from some private laboratories between \u00a0\u00a02016 to 2018. All patients were diagnosed by pathologists according to conventional methods that involved cancer parameters and clinical symptoms. Samples were included 20 men and 17 women ages at a rate of 20 to 72 years. Patients were early detected (without treatment with chemotherapy or any drug).This study had included two part the histological study which carried out by using immun ohistochimstry technique, and molecular study which carried out by using European biomedicine (BIOMED-2) multiplex PCR and heteroduplex technique for clonality detection of Immunoglobulin heavy chain (IGH) in NHL and DNA extracted from formalin fixed paraffin-embedded\u00a0(FFPE) tissue. The results of immune histochimstry was revealed positive staining in cytoplasmic as a perinuclear of \u03b2 -catenin in tumor cell that was detected in 30\/37 (80%) case of non Hodgkin’s lymphoma and 7\/37(20%) that was negative for b-catenin, while 4\/19 (21%) case of normal lymphoid tissue were showing a positive staining outside the germinal center and in all case of normal lymphoid tissue showed the positive staining in germinal center. Nuclear \u03b2 -catenin was observed in 7\/37 (18%) cases of non-hodgkin’s lymphoma, while no positive staining in nuclear was shown in normal lymphoid tissue.\u00a0 Metadherin (MTDH) was detected as a positive in 24\/37 (64%) cases non hodgkin’s lymphoma, while 13\/37 (36%) cases were detected as a negative for MTDH.In normal lymphoid tissue, 15\/19 (79%) of cases also detected as a negative for MTDH and 4\/19(21%) of case was showed positive staining for MTDH in lymphocytes. In molecular study the positive clonality was detected in rate about 70% (26\/37) as a monoclonal while 29% (11\/37) showed a polyclonal pattern. In a group with Diffuse large B-cell lymphoma(DLBCL) clonality was observed in 66%(10\/15) as monoclonal. In small lymphocytic lymphoma(SLL),the monoclonality that was detected in 75%(3\/4). In Burkitt\u00a0lymphoma (BL) monoclonality was shown in 66 %(2\/3). In mucosa-associated lymphoid tissue lymphoma (MALT)type mono clonality was shown in 50%(1\/2) and in patients diagnosed with follicular Lymphoma(FL) and nodal marginal zone lymphoma(NMZL)showed 100% of case as a monoclonal pattern. In normal lymphoid tissue 100 %(19\/19) the clonality showed a polyclonal pattern. In conclusion, the nuclear \u03b2-catenin pathway may suggest as a therapeutic target for DLBCL and SLL. MTDH expression in NHL cells can be enhanced cell proliferation, which leads to inhibition the apoptosis of cells .MTDH was over expressed in NHL with nuclear Wnt\/\u03b2 -catenin expression, and\u00ad confirmed that MTDH worked as an oncogene to promote lymphoma cells proliferation and inhibit their apoptosis through wingless-type(Wnt) \u03b2-catenin signal pathway. The best kit was used in this study for DNA extraction from FFPE tissue was promega kit for total DNA. The procedure of a BIOMED-2 was used as a best way to clonality analysis of IGH gene rearrangements (framework I, II, III) in the diagnosis of NHL. According to above this study can conclude two main things.
