Thesis on Bacteriologica Study of Meningitis Patient in Children by Abbas Hameed Shakur and supervised by Asst.prof.Dr.Luma Abdulhadi Zuain<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">Summary<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">The present study including collected 248 clinical samples of cerebrospinal fluid (CSF) from patients, from the Child Protection Teaching Hospital in the Medical City Complex in Baghdad. For the period of July to October 2018. All isolates were identified depending on macroscopic, microscopic, biochemical tests and definite with Vitek-2 compact system. Forty two isolates were obtained\u00a0 from samples showed a growth in 42 samples .Sixteen\u00a0 isolates belongs to Staphylococcus spp. 38.1%.,the number of\u00a0 the percentage of according to species\u00a0 as follow:7\/16(43.75%) of S.epidermidis, 3\/16(18.75%) of S.hominis and S.haemolyticus,2\/16 (12.5%) of S.aureus, 1\/16(6.25%) of S.warneri. Whereas the Gram negative was 1(2.38%) isolate of Pseudomonas putida, Escherichia coli, Klebsiella pneumonia, and 2(4.76%) isolates, Burkholderia cepacia and Acinetobacter baumannii, and 19(45.24%) isolates were other. These isolates showed differences in susceptibility to antibiotics, the results showed all isolates were resistance to Benzylpenicillin 100%,while the resistance to Oxacillin were\u00a0 85.7%, Gentamicin 28.57%, Tobramycin 21.42%, Levofloxacin 28.57%, Moxifloxacin14.28%, Erythromycin85.7%, Clindamycin42.85%, Tetracycline28.57%, Fusidic Acid64.28%,Rifampicin35.71% and Trimethoprim28.57%.While they were sensitive 100% for Linezolid, Teicoplanin, Vancomycin, Tigecycline and Nitrafurantion. Four different antibiotic resistance patterns were observed for S. epidermidis, one pattern for S.aureus,two patterns\u00a0 forS.haemolyticusand one pattern forS. warneri<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">These isolates were tested for ability to biofilm formation by using Luria Broth and tryptone soy Broth supplemented with 0.25% and 2% glucose.The ability of the isolates to form biofilm with Luria broth,for S.epidermidis, S. aureus, S. haemolyticus, S.hominisandS. warneriwere7 \/ 7( 100%),1\/2(50%),2 \/ 3 (66.66%), 1\/3 (33.33%),1\/1 (100%)were not forming biofilm respectively.As for using a tryptone soya broth TSB supplemented with 0.25% sugar glucose,Bacterial isolates of S.epidermidis, S. aureus, S. haemolyticus and S.hominis were6\/7 (85.71%), 2\/2 (100%),2\/3 (66.66%) \u00a0and \u00a03\/3 (100%) were weakly biofilm\u00a0 formed respectively.When using TSB medium supplemented with 2% glucose, Staphylococcusspp. isolates were all 100% weakly biofilm formed. The study showed the effect of MIC and MBC of Erythromycin (8) \u03bcg\/mL and (16)\u03bcg\/mLagainst Staphylococcus spp. Constituent of the biofilm.In the current study, the antibiotic showed the inhibitory effect in the bacteria that forming the bio film. When studying the susceptibility of bacteria to adhesion using Polystyrene dishes, The numbers of adherent cells were S.epidermidis reached 230 cells \/ field, S.aureus had 3 cells \/ field, while S.haemolyticus has more than 300 cells \/ field, S.hominis bacteria had 62 cells\/fields, finally the number of S.warneri was 40 cell \/ field. As for when using a Vero cell line of epithelial cells. The results \u00a0of current study according to the cultivation of bacterial dilutions was in S.epidermidis were 9 cells at 1: 100 dilution. The number of S.aureus and S.haemolyticus adherent to epithelial cells was more than 300 cells,S.hominis were 30 cells. While the number of bacterial cells in S. warneriwas 292 cells and examined the ability of Staphylococcus spp. isolates to spreading. The results showed their mobility (motility) as well as giving different forms depending on the species \u00a0of bacteria, the spreading of S.pidermidis was 7.7-16.5 mm,S.aureus ranged between 8.97 -16.0 mm,S.haemolyticus ranged between 8.0-14.6 mm,S.hominis ranged between 10.6 &#8211; 16.3 mm\u00a0 and S.warneri reached 16.8 mmAs for molecular detection genes of the entire isolates contained eno gene, which encoded to (Laminin binding protein). The gene bap that encodes to Biofilm association protein (Bap), the results of our current study showed that none of the bacterial isolates of Staphylococcus spp.has the gene.
