      Biosynthesis of Zinc Oxide Nanoparticles by Pseudomonas aeruginosa Locally Isolated<\/span><\/h3>\n

      This study aimed to synthesize bio-zinc oxide nanoparticles from Pseudomonas aeruginosa bacteria from different sources and then study the effect of these synthetic particles on some virulence factors of Pseudomonas aeruginosa bacteria<\/span><\/h3>\n

      104 swab samples were collected from different clinical sources of different ages and for the period from 9\/12\/2018 to 3\/3\/2019 and from the Medical City Hospitals in Baghdad (Baghdad Teaching Hospital, Ghazi Hariri Hospital, Burns Hospital, and Educational Laboratories).After microscopic and biochemical tests, 58 isolates were obtained from Pseudomonas aeruginosa<\/span><\/h3>\n

      Vitek compact system 2 was used to determine the minimum inhibitory concentrations of thirteen antibiotics for 30 isolates, where the isolates were highly resistant to Colistin and Tobramycin at 53.3%, 16 isolate Colistinand 56.7% with 17 isolate to Tobramycin<\/span><\/h3>\n

      Then, the ability of Pseudomonas aeruginosa bacteria to develop virulence factors was detected. 18 out of thirty isolates were composed of the pyocyanin pigment.While all the isolates under investigation had the ability to replace blood type beta to alpha. When examining the ability of bacteria to produce protease enzyme, it was observed that 25 out of thirty isolates possessed the ability to form protease enzyme at a rate of diameters ranging from 12-20 mm.<\/span><\/h3>\n

      When isolating the ability of bacteria to produce iron carriers, all isolates were produced for iron carriers. In the study of the ability of bacteria to form a biofilm, two methods were used to detect the first using Congo red agar method the purpose of this test was to study the ability of bacteria to form adhesion cells. All the isolates were made up of this layer, but in different proportions, it was found that 53% of the bacteria were highly formed according to the possession of a dark black color after growth and 47% were medium composition.<\/span><\/h3>\n

      The second method was done using micro titer plate. The results showed that all the isolates under study had the ability to form the bio film to varying degrees compared to negative control where 14 samples were poorly formed, 12 were medium formation and four were highly formed.<\/span><\/h3>\n

      The most antibiotic-resistant isolation was then selected for use in the biosynthesis of zinc oxide nanoparticles. The appearance of a white precipitate at the bottom of the test flask is evidence of this transformation.<\/span><\/h3>\n

      The properties of the resulting nanomaterials were studied using diagnostic devices such as UV spectroscopy, atomic force microscope, X-ray diffraction, infrared spectroscopy, scanning electron microscopy and zeta potential .The results of these devices indicate the formation of zinc oxide particles in pure and different forms and sizes ranging from 20 to 100 nanometers.<\/span><\/h3>\n

      The activity of synthetic nanoparticles was tested against Pseudomonas aeruginosa bacteria, where three different concentrations (0.01, 0.1, and 0.5) mg \/ ml were used.Concentrations of 0.1 and 0.5 showed efficacy in inhibiting bacterial growth while 0.01 gave no activity.<\/span><\/h3>\n

      The ability of synthetic nanoparticles to inhibit some virulence factors of Pseudomonas aeruginosa bacteria was studied.The ability of zinc oxide nanoparticles to inhibit the production of biofilm was studied.<\/span><\/h3>\n

      All the biofilm-producing isolates were treated with two concentrations of nanomaterials (0.1, 0.5 mg \/ ml) separately. Where 100 ul of concentrations were added to 100 ul of bacterial suspension in the micro titer plates and then incubated for 24 hours and then complete the steps to read the results. The results showed that the inhibition rate at the concentration of 0.5 was the largest by 83.8%, while the inhibition at the concentration 0.1 was (80%).<\/span><\/h3>\n

      Eight isolates of several antibiotics resistant\u2019s were tested using nanoparticles to detect the ability of these molecules to inhibit the production of hemolysin.<\/span><\/h3>\n

      Two concentrations of 0.5 and 0.1 mg \/ mL were used the isolates were planted on the nanoparticle-containing blood medium, where it was observed at 0.5 concentration that all isolates lost their ability to produce beta hemolysin and became alpha-hemolysin-producing while 0.1 isolates remained able to form beta-hemolysin<\/span><\/h3>\n

      The effect of zinc oxide nanoparticles on the production of pyocyanin dye was studied for a group of isolates resistant to several antibiotics (eight isolates) of Pseudomonas aeruginosa bacteria. In Concentration 0.5 all isolates lost their ability to produce dyewhile in the concentration of 0.1 only two samples were able to dye loss.<\/span><\/h3>\n

      An experiment was conducted to detect the effect of zinc oxide nanoparticles on the production of protease enzyme for some isolates resistant to many antibiotics of Pseudomonas aeruginosa bacteria using two concentrations:<\/span><\/h3>\n

      0.5 And 0.1mg\/ml on the milk agar where it was observed at a concentration of 0.5 decreased the proportion of enzyme production while proteases were not affected at a concentration of 0.1 mg \/ ml.<\/span><\/h3>\n

      Finally, an experiment was carried out to detect the effect of zinc oxide nanoparticles on the production of Siderophores for some isolates resistant to many antibiotics of Pseudomonas aeruginosa bacteria using two concentrations of 0.5 and 0.1 mg \/ ml on the nutrient agar containing nanoparticles.It was observed that in both concentrates all the isolates used in the experiment had weakened their production of Siderophores<\/span><\/h3>\n","protected":false},"excerpt":{"rendered":"
