    Abstract<\/span><\/h3>\n

    In this study, (100) samples were collected for different pathological cases to investigate Pseudomonas aeruginosa where (41) isolates were diagnosed with initial diagnosis by biochemical tests and then diagnosed with Vitec – 2 compact device.They were then given an allergy test for six types of antibiotics.<\/span><\/h3>\n

    Lipopolysacharide was extracted from isolation (27) by hot phenol and the crude lipopolysaccharide was obtained from the aqueous phase. The extract was partially purified by gel filtration technique using (Sephadex – G200). The concentration of carbohydrates and proteins for crude extract and purified by chemical methods was estimated as (70.1%) and (29.8%) respectively for the crude extract, while the percentage in the purified extract was (77.4%) and (21.5%) for carbohydrates and proteins respectively. The enzymes (Dnase, Rnase and Protase) were not used in the extraction and purification processes. The molecular weight of the purified extract was estimated using gel-filtration technique using (Sephacryl S-200) gel. The lipopolysaccharide and its biologic activity were detected using the tool (Limulus amebocyte lysate).The treatment of purified LPS with the reagent resulted in a clot indicating the efficacy of the biologic extract. The effect of the heat factor on the extract was investigated, where the extract was treated by boiling using a boiling water bath at a temperature of (100) \u00b0 C within a concentration of (200) g\/ml , period of time (5,10,15) minutes and then detected using the reagent (LAL) , where the extract showed resistance to thermal ranges used and within the specified concentration and led to be a clot when treated with the reagent. The effect of moist heat of the extract was investigated by using the sterilizer at (121)\u00b0C and pressure (15) psi at afor (5,10,15) minutes with concentration of (200)g\/ml ,It was then detected using a detector (LAL) and the extract’s resistance to moist heat was determined within the specified concentration and time intervals. The effect of\u00a0 partially purified extract on Candida glabrata yeast was studied using microdilution method. The results showed that partially purified extract had inhibitory effect on Candida glabratayeast by (45%) . Then the heat treated extract was used to investigate it,s effect on Candida glabratayeast and at the same concentrations and durations used with the (LAL) reagent.The results showed that the boiling heat and moist heat did not affect the efficiency of inhibitory activity of purified LPS on Candida glabrata and the inhibitory ratios were comparable to the effect of non-treated partially purified lipopolysaccharide.<\/span><\/h3>\n
