FAYIDH<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">Abstract<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">From 120 patient samples obtained from some hospitals in Baghdad, during the period from 18\/10\/2018 to 8\/1\/2019, 43 isolates belonging to the genus S. aureus were diagnosed, based on traditional diagnostic methods, then diagnosed isolates using Vitek device -2 Compact system<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">The sensitivity of the 43 isolates to 15 antibiotics due to different antibiotic family were tested using the diffusion disc tablet method, the highest resistance was recorded for the\u00a0 Methicillin ME 95.3%, whereas the resistance to Vancomycin was non-existent, while the percentage of resistance to Oxacillin (OX) was 72%, Gentamicin (CN) (55.8%), Erythromycin(E)\u00a0 (37.2%), Azithromycin (AZM) (34.8%), Trimethoprim TMP (30.2%), Tetracycline (TE) (27.9%), and other antibiotics showed low resistance percentages<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">To confirm the sensitivity test by tablets, the Vitek device was used to measure the values of the minimum inhibitor concentration (MIC). The results of this method were exactly the same as the results of the method of spreading using tablets in all antidotes, except Trimethoprim, where two isolates showed their resistance to the method of tablets while all isolates were sensitive when using Vitek-2. The cause may be the heterogeneity of the antibiotic concentration used in both methods, or that the components of the antibiotic tablet are somewhat different from those available in Vitek-2<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">The susceptibility of the 43 local bacterial isolates of S. aureus to the production of broad-spectrum beta-lactamase enzymes was studied. The test results showed that only 14 isolates (32.5% of the total isolates) were able to produce broad spectrum beta-lactamase enzymes. The rest of the isolates did not show a positive result, which is the overlap of the inhibition areas with each other<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">As for the possession of 43 bacterial isolates of the Efflux pumps that reflect the efficiency of resistance to antibiotics, it was revealed in morphologically using the Cartwheel Method with the dye Ethidium bromide-Agar, (CW ETBr) Ethidium Bromide. The results showed that all isolates of the study, gave positive results through their brightness varied in the concentration of 0.5\u00b5 \/ ml, and they were also radiant in all concentrations indicating the power of the Efflux pumps they possess<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">The molecular study included three main axes<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">The first axis included the molecular diagnosis of the study isolates using the 16S rRNA gene primers. The results confirmed that all the selected isolates were previously classified based on their phenotypic characteristics and biochemical efficacy, as well as the results of Gp- Vitek-2 as genetically representative S.aureus<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">The second axis included the detection of antibiotic resistance genes in all isolates. They were represented by the use of special primers that encode the gene mecA resistance gene. The results showed that all isolates possessed the mecA gene, except for two isolates that did not possess this gene. The genes of resistance to erythromycin were also detected in 16 erythromycin-resistant isolates using three special precursors for genes that encode the erythromycin-resistant characteristic of ermC, ermB and ermA. Results show that ermA gene exists in only one isolation, while the ermB gene was found in 14 isolates (87.5%), and the ermC gene appeared in 11 isolates (68.75%) of the total erythromycin resistance<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">The third axis included the detection of genes responsible for the isolation of Efflux pumps, represented by the use of special gene primers, norA gene. norB gene The results of genetic detection showed that all isolates (43) possessed the norA gene, while the norB gene in (38) isolation only<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">To determine the sequence type and shape of the sequences in the local isolates under study, four isolates were selected for sequencing 16S rRNA, as well as sequencing of ermA, ermB and ermC genes<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">Initial results recorded percentages of differences with the global isolates preserved in the GeneBank of NCBI, as well as a number of mutations of replacement,\u00a0 and deletion in the sequence of nitrogenous bases in the gene DNA of elected local isolates. This encourages their registration as distinct local samples at GeneBank of NCBI, as well as further studies and research for other local isolates<\/span><\/h3>\n<h3 style=\"text-align: left\"><span style=\"color: #993366\">The results of this study confirm the prevalence of many bacterial strains of S. aureus in Iraq, varying in biochemical characteristics, as well as their resistance to antibiotics, as well as genetically different.
