Disease (IBD) is a chronic ,multifactorial disease which include two major types :ulcerative colitis( UC) which infects the colon in large intestine and crohn&#8217;s disease (CD) which can affects any part of the digestive tract from the mouth to anus ,Although the causes of the disease did not identified exactly but the researches proved that genetic environmental factors leading to an abnormal immune response for mucous layer of the bowel (mucosa) towards antigens with in the intestinal Lumen. Current study aimed to investigate genetic polymorphisms inSOD1and TNF-\u03b1 genes in some Iraqi patients with IBD because of its\u00a0 great importance in the occurrence of this disease and its association with occurrence of tumors and cancers in gastrointestinal tract (GIT).This study included collection of blood samples from the patients who attending gastrointestinal and liver disease in the city medicine from September to December .Genetic polymorphisms were investigated for each SOD1 and TNF-\u03b1 genes by Restriction Fragment Length Polymorphisms(RFLP) and Sequencing. Current study include 75 samples from IBD Iraqi patients after confirming the diagnosis of the disease by the specialist doctor ,and 20 samples took from healthy as a control group (HC) for comparison purposes, We kept samples in fridge until use .The results showed that there was a high significant depending on some clinical characteristics of patients ,The incidence rate was 43(57%),32(43%) for females and males respectively as well as The incidence rate of Ulcerative Colitis UC (62.7%) was high compared with Crohn&#8217;s disease CD(37.3%) with high significant. The results of gel electrophoresis of DNA fragments for SOD1 gene which amplified by PCR technique and cut by restriction enzyme Hhal for Homozygous A Genotype gave one fragment size (278bp) and the ratio of Homozygous A Genotype in IBD Patients (94.67%) compared with healthy group (75%)( p=0.0093,Chi=7.149**,OR=1.095) with significant. The results of gel electrophoresis of DNA fragments for SOD1gene for Heterozygous A&gt;C Genotype showed different size of fragments(278,207,71bp)\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 Heterozygous A&gt;C genotype ratio (5.33%) in IBD\u00a0 patients lower than healthy group (HC) (25%) with significant(p=0.0093, Chi=7.149**, OR=1.095)the Homozygous C Genotype not appear in both patients and healthy group.The results of allele frequency for A allele in IBD patients and healthy group respectively (97%,87.5%),and C allele in IBD patients and healthy group respectively (3%,12.5%). In addition, The results of gel electrophoresis on Agarose gel for DNA fractions for TNF-\u03b1\u00a0 -1031 gene which amplified by PCR technique and cut by restriction enzyme BbSI :There are different size of DNA fragments for Homozygous T Genotype size(251,13bp) that indicate to existence different genotype for the TNF-\u03b1\u00a0 -1031\u00a0 gene ,the results showed that the Homozygous T Genotype in IBD patients (62.67%) compared with the control group (Healthy) (HC 70%)(p=0.061, ChI=3.091 NS, OR=0.377),The results of gel electrophoresis on Agarose gel for DNA fraction for TNF-\u03b1\u00a0 -1031 \u00a0for Homozygous C Genotype appear different size of DNA fragments(180,71bp) and the ratio of Homozygous C Genotype in IBD Patients (5.33%) compared with healthy group (5% )(p= 0.872, ChI=0.055 NS, OR=0.0162), The results of gel electrophoresis on Agarose gel for DNA fraction for The Heterozygous Genotype\u00a0 T&lt;C appear different size of DNA fragments(251,180,71bp), The Heterozygous Genotype\u00a0 T&lt;C ratio in IBD Patients(32%) compared with HC group(25%)( P=0.069, ChI= 2.983, OR=0.352).The results of allele frequency based on Hardey Weinberg showed very high ratio for T allele (79%) compared to C allele (21%) in patients while the results\u00a0 in healthy group for T allele(83%) and C allele (17%) with non significant ,Current study revealed the active role of Homozygous A Genotype in the occurrence of the inflammations where the value of cause active agents was(EF=0.362,OR=1.095) while the Heterozygous Genotype A&gt;C play protective role in block the incidence the value S (PF=0.176,OR=1.095) The value of PF for Heterozygous Genotype CC was 0.116 , As indicated for the active role of Homozygous T Genotype in emergence of inflammations the value of( EF=0.511,OR=0.377) on the contrary of that Homozygous C Genotype and Heterozygous Genotype T&gt;C possess protective role against disease (PF=0.152,0.376) for CC and CT geno type respectively and the values of (OR=0.0162,0.376) for CC and CT genotype respectively\u00a0 .In conclusion the results showed that the incidence of UC higher than CD and the incidence of IBD in ages less than 50 years higher than 50 or more years old .As well as in female more incidence than in male with high significant differences(p&gt;0.01),As we found high percentage of homozygous allele AA for SOD1 gene compared with homozygous allele CC and heterozygous allele AC significant differences, and the high percentage of homozygous allele TT for TNF-\u03b1 -1031 gene compared with
