    Molecular Detection of some resistance Geneses of Klebsiella pneumoniae isolated from Clinical Samples<\/span><\/h3>\n

    Abstract<\/span><\/h3>\n

    \u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 In this study, a total of 381 clinical samples included 96 burns samples, 47 wounds samples, 67 sputum samples, 92 urinary tract infections samples, 11 otitis media samples, 63 stool samples and 5 cerebrospinal samples were collected from five hospitals in Baghdad city, during a period from 23\/7\/2018 to15 \/1\/2019. The samples were cultured on MacConkey agar and blood agar media to isolate Klebsiella pneumoniae bacteria. The study reached for the following<\/span><\/h3>\n

    The results showed that rate of K. pneumoniae isolates was 89.8% (342\/381) of total clinical samples and, it was found that 11.1 % (38\/342) of K .pneumonia isolates were resistant to carbapenem (CRKP). These CRKP isolates was subjected to study<\/span><\/h3>\n

    The results of phenotypic betalactamases detection showed that all CRKP isolates (100%) did not produce extended spectrum betalactamases\u00a0\u00a0 using double disk synergy test, while all these isolates were 100% produced metallo-betalactamases (MBLs) using imipenem-EDTA combined disk test<\/span><\/h3>\n

    Result of antibiotic sensitivity test for CRKP isolates against 15 antibiotics using agar disk diffusion method showed\u00a0 that sensitive rate were 65.8% and 28.9% for levofloxacin\u00a0 and\u00a0 ciprofloxacin respectively, while all CRKP isolates were 100% resistant (sensitivity 0%) for the other 13 antibiotics: oxacillin,ceftriaxone, amoxicillin-clavulanate, piperacillin, piperacillin \u2013tazobactam,cefotaxime, cefepime, ceftazidime, aztreonam, tobramycin, amikacin,\u00a0 gentamicin and trimethoprim-sulfamethoxazole. The resistance results of metallo-betalactamase producing CRKP isolates for antibiotics according to MICs values were\u00a0 agreement to results that obtained by disk diffusion method<\/span><\/h3>\n

    The result of molecular detection of blaKPC-2,blaNDM-1 and bkaOXA-48 genes responsible partial resistance for carbapenems antibiotics\u00a0 showed that 100%, 84.2% and 65,7%\u00a0 of CRKP isolates were contained the genes respectively,\u00a0 while the blaIMP and blaVIMgenes were\u00a0 not recorded (0%) in all CRKP bacterial isolates<\/span><\/h3>\n

    The results of bio film formation test of\u00a0 CRKP isolates by microtiter plate method at wave length 630 nm showed that all isolates (100%) was formed it, 5.3% and 94.7% of bacterial isolates was formed moderate bio film\u00a0 and weak bio film respectively.<\/span><\/h3>\n

    The results of side rophores production test of CRKP isolates showed that all isolates (100%) was produced it.<\/span><\/h3>\n

    The results of efflux pumps production test of CRKP isolates showed that all isolates (%100) was production it.<\/span><\/h3>\n

    The sequencing analysis for 16S-23SrDNA gene (one CRKP isolates), four isolates for each of blaKPC-2, blaNDM-1 and blaOXA-48 genes were done and the results appeared (38) mutations for each blaOXA-48 and blaNDM-1 and one mutation for each 16S-23SrDNA and blaKPC-2.<\/span><\/h3>\n
