            <\/h4>\n

            The study includes the development of two new spectrophotometric methods for the determination of thiamine hydrochloride (Thia.HCl) and folic acid(FA) individually and simultaneously in their pure form and in pharmaceutical preparations. The thesis is divided into three chapters.<\/span><\/h4>\n

            Chapter one<\/u><\/span><\/h4>\n

            This chapter includes an overview of the UV-vis spectrophotometry and a brief on the properties of Thia.HCl, FA, and the 4-amino-antioxidant reagent (4-AAP). Moreover, a light was thrown on the general bases of diazotization reaction and its importance and application in analytical chemistry in addition to a survey on different analytical methods mentioned in the literature concerning the determination of the two drugs under study.<\/span><\/h4>\n

            This chapter also includes a detailed explanation of two different approaches that have been relied upon to reach the optimum experimental conditions, namely the univariate and multivariate methods (experiment design). Also in this chapter, an idea was given about the derivative spectrophotometry that was relied upon in the simultaneous determination of the two cited drugs and the aim of the study was mentioned.<\/span><\/h4>\n

            Chapter two<\/u><\/span><\/h4>\n

            The chapter contains a detailed description of spectrophotometric methods based on the diazotization and coupling reactions to form a stable azo dye and its application in the felid of spectrophotometric determination of the two investigated drugs. This chapter deals with the practical part that includes a description of the chemicals and instruments used and the preparation of the solutions of the used reagents and the standards of the two drugs along with the section for the results and discussion.<\/span><\/h4>\n

            The study showed that the developed spectrophotometric method for the determination of Thia.HCl depends on its coupling with the diazotized 4-AAP (prepared in the presence of hydrochloric acid and sodium nitrite) in a basic aqueous medium to yield a dark yellow water-soluble azo-dye whose absorption is measured at a wavelength (\u03bbmax<\/sub>= 449 nm). Two methods were followed to attain the optimum conditions affecting the color-forming reactions, namely the traditional univariate and a chemometric multivariate based on the experimental design. It was found through the results obtained under the optimum conditions and the analytical characteristics and statistical data of calibration graphs for the determination Thia.HCl. that the proposed method is reliable, as the value of the molar absorptivity is equaled to 6742.26 L. mol-1<\/sup>.cm-1 <\/sup>and the Sandal’s index was equal to (0.05) \u03bcg.cm-2<\/sup>.\u00a0 Beer\u2019s law is obeyed in the concentration range of (5-60) \u03bcg.mL-1 <\/sup>of the drug, the recovery percent ranged between (95% -101%) and the relative standard deviation percent was less than 1%, while the values of limit of detection (LOD) and limit of quantification (LOQ) were 0.0822 \u03bcg.mL-1 <\/sup>and 0.2739 \u03bcg.mL-1 <\/sup>respectively. After it was confirmed that the presence of common drug additives (excipients) did not interfere, even when they are present in large quantities, the method was successfully applied to pharmaceutical products containing Thia.HCl using the direct and standard addition methods.<\/span><\/h4>\n

            On the other hand, the developed spectrophotometric method for the determination of FA\u00a0 based on the quantitative diazotization of the drug (in the presence of sodium nitrite and sulfuric acid)\u00a0 and the resulting diazonium salt was coupled with 4-AAP in basic medium. A water-soluble orange colored dye was formed which shows maximum absorption at (\u03bbmax<\/sub>= 449 nm ) versus blank. Under optimum conditions, Bear’s law was applicable in the rang of FA concentration (2-100) \u03bcg.mL-1 <\/sup>\u00a0with a value of molar absorptivity of 3266.36 L.mol-1<\/sup>.cm-1 <\/sup>and the Sandal’s index was equal to (0.1352) \u03bcg.cm-2<\/sup>.\u00a0 The percent of recovery of the developed method was in the range of\u00a0 (97-102)%\u00a0 and the value of relative error percent was less than 1%, while the values of LOD and LOQ were 0.1482 \u03bcg.mL-1 <\/sup>and 0.4941\u03bcg.mL-1 <\/sup>respectively. The developed method showed no significant interference when common excipients are present, hence it was successfully applied to the determination of the drug FA in some of its pharmaceutical preparations.<\/span><\/h4>\n

            Chapter three<\/u><\/span><\/h4>\n

            This chapter deals with the possibility of using derivative spectrophotometry for the simultaneous determination of Thia.HCl and FA in their binary mixture ( i.e. a mixture of their pure forms or of their pharmaceutical preparations) without using chromogenic reagents by the application of first, second and third derivatives techniques. The chapter also includes the experimental practical part which deals with mentioning the used instruments, chemicals used and methods for their preparations.<\/span><\/h4>\n

            The results of the first derivative show that the zero cross measurements carried out at different approparaite wavelengths are suitable for individual and the simultaneous determination of the studied drugs. It was found that the value of the derivative spectrum height above the baseline at the zero cross of either drug is directly proportional to the concentration of the other drug. Therefore, wavelengths 283.4 nm and 266.9 nm (the zero-cross of FA) were used for the determination of Thia.HCl in the presence of FA and in the same manner a wavelength of 309 nm (the zero-cross of Thia.HCl) was chosen to determine FA in the presence of Thia.HCl. Furthermore, when the measurements were carried out on the second derivative spectrum of the binary mixture,\u00a0 the two wavelengths 274.6 nm and 327.3 nm were selected for the determination of Thia.HCl and FA respectively. For the third mode derivative, the wavelengths 293.6 nm and 262.0 were chosen to determine Thia.HCl and FA respectively.<\/span><\/h4>\n

            The linearity ranges with respect to the two-drug concentrations for all derivative modes was (1-25) \u03bcg.mL-1<\/sup>. The values \u200b\u200bof the relative standard deviation percent (RSD%) and the relative error percent (RE%) for the results, obtained by using the three mods of derivative, ranged between (0.1436-4.6992) % and (-3.3389-2.1858) % respectively for Thia.HCl and (0.2040-2.8523) % and (-3.1977-3.6873) % respectively for FA when the two drugs were individually or simultaneously determined. Whereas, the values \u200b\u200bof RSD% and RE% ranged between (0.2040-3.5668)% and (95.8423-104.2677) % respectively for the determination of Thia.HCl and (0.2040-3.5668) % and (95.1475-104.9093)% respectively for the determination of FA in their pharmaceutical preparations.
