      . Using oxfendazole alone or in combination with praziquantel and Albendazole, and comparing them as an attempt to treat infected white mice experimentally<\/span><\/h3>\n

      \u00a0Evaluation of some hematological, biochemical and immunological parameters in the sera of infected mice during the stages of growth and development of Hydatid cyst, and determining the therapeutic efficacy of the drugs used in the study<\/span><\/h3>\n

      . A histopathological study to determine the histological change on sections of the liver and spleen of experiment mice<\/span><\/h3>\n

      .A histochemical Immunohistochemical study by detecting the level of histological expression of some of the apoptosis proteins Bcl2, Bax, Caspase 3, PD-1 and its linkage PD-L1 as a sign of the cellular death process programmed in the liver and spleen sections, and estimating the Level of alpha-necrosis factor (TNF-\u03b1) and interleukin-4 .<\/span><\/h3>\n

      The current research is deals with fundamental aspect of the mechanism programmed cell death, as it is one of the important steps in determining the therapeutic drug. Therefore, the effect of infection and treatment of hydatid cysts was highlighted to 90 white male mice Balb\/ c strain at the age of 4-6 weeks and divided into six groups (15 mice per group) experimentally infection with secondary hydatid cyst through I\/P\u00a0\u00a0 of 2000\u00b15 viability protoscolices and left the sixth group as a negative control group.\u00a0 Four months after infection four groups were treated for two months with three drugs which belong to benzimidazole\u00a0 and isoquinolineviz Oxfendazole (OXF 30 mg\/kg), Paraziquental (PZQ 40 mg\/kg) and Albendazole (ABZ 10 mg \/kg), drugs given to mice groups in single dose or mixed, as follows groups: OXF, OXF + PZQ, OXF + ABZ, and ABZ + PZQ.<\/span><\/h3>\n

      The study included several aspects<\/span><\/h3>\n

      First aspect: A study of some hematological and immunological parameters which included blood cell picture, delayed type hypersensitivity and index phagocytosis. Regarding\u00a0\u00a0 changes in differential white blood cell count, mice treated groups showed decreased percentages of eosinophil’s and lymphocytes with increased neutrophil cells and monocyte varied rate compared with positive control after the first and second month after treatment. The OXF and OXF + PZQ groups recorded a decrease in the total count of white blood cells compared to the positive and negative control group, while OXF showed an increase in both PCV and Hb also an increase in left foot pad thickness (0.77 mm) after 24 hours compared with other groups. As for the phagocytosis factor, the highest significant increase was in the OXF + PZQ and OXF groups with ratios of (69.36%) and (55.6%) respectively compared to other groups\u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0<\/span><\/h3>\n

      The second aspect of the study was to evaluate the effect of infection with hydatid cyst and different chemical treatment on liver functions where the study showed that liver enzymes such as GPT, GOT and ALP, as well as total protein, albumin and globulin serum level in mice treated with OXF alone and OXF + PZQ were closer to those of the negative group compared with the groups treated with OXF + ABZ and ABZ + PZQ.\u00a0\u00a0<\/span><\/h3>\n

      Third aspect: Evaluation of the criteria taken into consideration to determine the activity of the drugs used in this study were the reduction in weight and diameter of the cysts and the morphological changes occurred within the cysts weight and organ index of the liver and spleen\u00a0\u00a0 to calculated the relative therapeutic efficacy, as the highest treatment efficacy was found in the OXF + PZQ group at 91.08% and the lowest in the ABZ + PZQ group at 79.60%\u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0 \u00a0<\/span><\/h3>\n

      The fourth aspect of the study also included the evaluation of the pathological changes of liver and spleen of mice after treatment, the results indicated that the drugs used destroyed both the laminated\u00a0 and germinal layers of the hydatid cysts wall through exerting changes in comparison with positive control group, Positive control mice that exhibited histological changes as a result of hydatid cysts led to a loss of their natural structure<\/span><\/h3>\n

      The fifth aspect study\u00a0 \u00a0when investigating the role effect of infection and chemotherapy of hydatidosis on expression of some apoptotic markers in both liver and spleen through the Immunohistochemical (IHC) technique. Bax protein expression\u00a0\u00a0 was still elevated in liver tissue under treatment with OXF alone or in combination ABZ + PZQ, OXF + PZQ but it was reduced in spleen tissue under treatment with OXF in combination OXF + ABZ, ABZ + PZQ. Despite the little variation in the reduction of Bcl-2 protein among treated groups OXF+ABZ showed the lowest group in liver tissue while OXF+PZQ was recorded as lowest group of reduction in spleen tissue<\/span><\/h3>\n

      Caspase -3 protein expression also reduced after two months of treatment in combination treatments in comparison with non-treated group. The results showed strongly suggesting that restoration of inflammatory processes after reduction of PD-1\/PD-L1axis after treatment,\u00a0 the results showed that there were statistical significant reduction of PD-1\/PD-L1 expression in liver and\u00a0 spleen treated\u00a0 groups than those of none treated group\u00a0 \u00a0<\/span><\/h3>\n

      The study also included the role of some cellular interleukins in the immune response against hydatid cyst through the IHC, the statistical analysis showed the presence of high significant differences (p< 0.001) in the level of expression protein Tumor necrosis factor-alpha (TNF-\u03b1) and IL-4 in both liver and spleen tissue\u00a0 in compared with negative and positive control groups , reduction level of cytokines TNF-\u03b1 and IL-4 in mice treated with the four treated groups\u00a0<\/span><\/h3>\n

      This is the first study to describe the cellular expression of some apoptotic markers in the liver and spleen of mice treated with different anthelminthic drugs regimens after experimental cystic<\/span><\/h3>\n
