        Abstract<\/span><\/h3>\n

        \u00a0The current study aimed to determine the activity of regulatory T cells (Treg) in Iraqi patients with Rheumatoid Arthritis and patients with Psoriasis through evaluating the level of cytokines (TGF-\u03b21, IL-6) and gene expression of Leucine rich repeat containing 32(LRRC32 gene)(GARP) andIkaros Zinc Finger 4(IKZF4 gene) (EOS).The present study included 52 patients (females and males),\u00a0 30\u00a0 patients diagnosed with Rheumatoid arthritis(RA) and 22\u00a0 patients with psoriasis; their age ranged between (18-72) years, in Baghdad Teaching Hospital\/Rheumatology consult, Dermatology consult\/Medical City\/Baghdad. The control group consists of 24 samples, over a period from (August to November) 2019. The RA patients divided into 3 groups: the first group consisted of 7 patients who treated with biological therapy anti-TNF (Etanercept); the second group consisted of 9 patients who treated with chemotherapy MTX (Methotrexate), and the last group consisted of 14 patients who treated with bio chemotherapy, while the Psoriasis patients included 2 groups, the first one consisted of \u00a07 patients who treated with topical therapy (Salicylic acid, Betnosam, Dermovate and Vaseline) and the second group consisted of 15 patients who treated with biological therapy (Etanercept). The level of cytokines was measured by Enzyme-linked immunosorbent assay (ELISA) techniquewhile the gene expression level of\u00a0LRRC32\u00a0gene and\u00a0IKZF4\u00a0gene was measured by RT-qPCR technique<\/span><\/h3>\n

        The results of our study showed a significant decreasing (P \u2264 0.05) in the level of TGF-\u03b21 in the serum of RA patients thattreated with biological therapy\u00a0compared to patients that treated with chemotherapy and others treated with bio chemotherapy, as well as to control group, the levels of TGF-\u03b21 were (0.26\u00b10.05,0.55\u00b10.04,0.51\u00b10.06) pg\/ml respectively, while the level in control was (0.60\u00b10.05) pg\/ml<\/span><\/h3>\n

        The results also showed there was a significant decreasing (P \u2264 0.05) in TGF-\u03b21 serum level of Psoriasis patients that treated with topical therapy and others treated with biological therapy compared to the control group, where the TGF-\u03b21 levels were (0.31\u00b10.05,0.40\u00b10.06) pg\/ml respectively, while in control group was (0.60\u00b10.05) pg\/ml<\/span><\/h3>\n

        \u00a0 \u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 The results of the current study showed a significant increasing in the level of IL-6 in patients with RA that treated with chemotherapy and those treated with bio chemotherapycompared to control group (10.86 \u00b1 1.45, 12.03 \u00b11.97,7.60 \u00b1 0.67) pg\/ml respectively; alsothe results showed a non-significant differences in patients treated with biological therapy (11.14\u00b12.98) pg\/ml compared to control group (7.60 \u00b1 0.67) pg\/ml<\/span><\/h3>\n

        \u00a0 \u00a0 \u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 \u00a0In psoriasis patients, the results showed a non-significant differences in the level of IL-6 between patients groups (treated with topical therapy + treated with biological therapy) as compared with control;the IL-6 levels were (9.25\u00b1 1.62,7.17 \u00b1 0.63) pg\/ml respectively and its level in healthy group was (7.60 \u00b10.67) pg\/ml.<\/span><\/h3>\n

        \u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 The results of\u00a0LRRC32\u00a0gene expression\u00a0showed a higher significant percentage of the gene expression folding(1851.9%) in patients with RA whilethe percentage of control 100% . According to the type of therapy, the results showed a higher significant percentage of the gene expression folding of\u00a0LRRC32\u00a0gene\u00a0in the patients under biological therapy and others treated with bio chemotherapy (2256.7%, 1939.4%) respectively as compared to control percentage 100%, while there were a non-significant differences in patients treated with chemotherapy( 1400.9%) as compared to control percentage 100%.In psoriasis patients, the results showed a higher significant percentage of the gene expression folding of\u00a0the LRRC32\u00a0gene\u00a0(1030.7%) as compared to control percentage 100%,also there was a significant increasing in the percentage of the gene expression folding of\u00a0LRRC32\u00a0gene in patients treated with biological therapy and in patients treated with topical therapy (1192.3%, 682.6%) respectively as compared to control percentage 100%.<\/span><\/h3>\n

        \u00a0 \u00a0 \u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 The results of IKZF4\u00a0 (EOS) gene expression level in RA patients illustrate a higher significant percentage of the gene expression foldingof\u00a0IKZF4\u00a0gene\u00a0(992.1%)as compared to control percentage 100%.Also, the results observed a significant increasing in the percentage of the IKZF4\u00a0(EOS)\u00a0gene expression foldingin RA patients treated with biological therapy, chemotherapy, and patients treated with bio chemotherapy (1382.3%,998%,794.1%) respectively, as compared to\u00a0 control percentage 100% .<\/span><\/h3>\n

        \u00a0 \u00a0 \u00a0 \u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 Where the results illustrate a higher significant percentage of the IKZF4 gene expression folding in patients with Psoriasis (702.9%) as compared to control percentage 100%. According to the type of therapy, there was a higher significant percentage of theIKZF4 gene expression folding in patients treated with biological therapy (747%) as compared to control percentage 100%,while there were a non-significant differences between patients treated with topical therapy(607.8%) and patients treated with biological therapy as well as to control group.<\/span><\/h3>\n

        The results illustrated a significant negative coefficient correlation in this study (P \u2264 0.05) between\u00a0the LRRC32 gene\u00a0and TGF-\u03b21 levels in patients with RA and treated with biological therapy (r=\u00a0-0.426).
