In Iraq, Breast cancer is considered the most common cancer in women compared to other type of cancer<\/span><\/h3>\n

    \u00a0\u00a0\u00a0\u00a0 The current study aimed to evaluate the etiological role of the inhibitory check point molecules CTLA-4 and IL-10 in the tumoregenicity of breast cancer \u00a0,so this study included two parts. The first is serological study which was carried out by using Elisa technique to determine the serum level of IL-10 and sCTLA-4 level, while the second part was conducted by using PCR-RFLP to determine the genotyping\u00a0 polymorphism of CTLA-4 gene for the region\u00a0 +49 in exon 1 SNP(rs 231775 A\/G).and DNA sequencing assay to determine the genotyping\u00a0 polymorphism of CTLA-4 gene for the promoter region 318 SNP(rsrs5742909 C\/T)<\/span><\/h3>\n

    \u00a0\u00a0\u00a0\u00a0 Ninety\u00a0 peripheral blood samples were collected (45) pre-treatment (PT) patient women diagnose with breast cancer from the Oncology Teaching Hospital of the Medical City, and the rest (45) for apparently healthy women as control group .patient and control with\u00a0 age range (30-70) years. During the period from September 2019 to February 2020<\/span><\/h3>\n

    \u00a0\u00a0\u00a0\u00a0 The results of serological study demonstrated that, the IL-10 serum level of\u00a0 patients group was (56.49 \u00b1 18.32pg\/ml),it was elevated in patient serum as compared to the low level in control group (21.19 \u00b1 5.16pg\/ml) but there was a non-significant dissimilarity under (p>0.05).sCTLA-4 serum level of\u00a0 patient was (3.17 \u00b1 0.82 ng\/ml) with an elevation level of CTLA.4 in patient serum compared to control group(2.72 \u00b1 0.29ng\/ml), also with\u00a0 a non-significant difference (P>0.05)<\/span><\/h3>\n

    \u00a0\u00a0\u00a0 In regard to molecular genetics study,CTLA-4SNP (rs231775A\/G) it has been observed that there was three genotypes (AA, AG, GG),AA genotype recorded a high frequency in control group (95.56%) versus patients (64.44%)compared to the other genotype AG, with a high significant difference according to Fisher\u2019s exact probability. Serum level of sCTLA-4 in patient IDC with genotype AA was high with a non-significant difference, while in patients ILC, the genotype AA recorded a high significant difference as compared to control and patients with AG,GG genotypes<\/span><\/h3>\n

    \u00a0\u00a0\u00a0\u00a0\u00a0 DNA nucleotides sequencing was identified in the promoter area of CTLA-4 gene. SNP (rs5742909 C\/T) which has three genotypes (CC, CG,CT) while the other genotypes(GG,GT,TT) was absent in 18 breast cancer patients and 11 control. The homozygous genotype CC recorded was a high (66.67%) in patients versus the control (45.46%) with a non-significant dissimilarity (P>0.05).The genotype CG was (22.22%) in patients, it was high in patients versus control (18.18%) and it showed a non-significant difference under (P>0.05) according to Fisher\u2019s exact probability. While the genotype CT which was recorded (11.11%)in patients, it was low in patients versus control (36.36%) and it showed a non-significant difference under (P>0.05) according to Fisher\u2019s exact probability. So according to the present results we can conclude that the genotypes AG in CTLA-4SNP (rs231775A\/G)\u00a0 and CC and CG in CTLA-4 gene SNP (rs5742909 C\/T) were represented by the etiological fraction and the common genotype in Iraqi females the genotype AA in CTLA-4 SNP (rs231775A\/G) and the genotype CC in CTLA-4gene SNP (rs5742909 C\/T) . The most common alleles in the study group was\u00a0 allele A in CTLA-4 SNP (rs231775A\/G) and\u00a0 allele C in CTLA-4 gene SNP (rs5742909 C\/T) , which may lead to increase the susceptibility of Iraqi female to have breast cancer. (The SNPs were registered in global database)<\/span><\/h3>\n

    Recommendations<\/span><\/h3>\n

    -Determine the serum level of IL-10 in the same patients before and after mastectomy or resection surgery and then after treated with chemotherapy and explain the results.<\/span><\/h3>\n

    – Study the role of IL-10 and sCTLA-4 in breast cancer patients by using real time PCR to study the levels of gene expression and compare the results.<\/span><\/h3>\n

    – Further studies about other CTLA-4 gene SNPs should conduct in order to shed a light on the relationship between polymorphism and susceptibility for breast cancer<\/span><\/h3>\n
