      Knowing the effect of acetic acid and the amino acid Tryptophan in both its isomeric forms (D and L) as an inhibitor of biofilm formation in P. aeruginosa<\/span><\/h3>\n

      Abstract<\/span><\/h3>\n

      In the current study, 100 samples were collected from different clinical sources which included: 26 burns samples, 25 wounds samples, 26 urine samples, 12 sputum samples, 9 blood samples and 2 middle ears from Baghdad Teaching Hospital, Burns Specialized Hospital, Shahid Ghazi Hariri Hospital for Specialized Surgery, the National Center for Teaching Laboratories, the Central Child Teaching Hospital and the Al-Imamayn Al-Kadhimin (Peace be upon him) Medical City for the period from 30\/9\/2020 to 1\/1\/2021<\/span><\/h3>\n

      After performing microscopic and biochemical examination, 53 isolates belonging to Pseudomonas aeruginosa were obtained. The minimum inhibitory concentration ( MIC ) value of twelve antibiotics was determined using the Vitek2 compact\u00a0 system\u00a0 for forty-five isolates, as 33 isolates were resistant to Imipenem at a rate of 73.3%\u00a0 while 38 isolates were sensitive to the antibiotic at a rate of 84.5%.<\/span><\/h3>\n

      The ability of bacterial isolates to be tested on blood analysis was revealed, the results showed that all isolates produced the enzyme Hemolysin at a rate of 100% and degradation of beta type, and the ability of isolates to form biofilm was revealed using the microtiter plate method\u00a0 and the results showed that 36 Out of the 45 isolates have the ability to form a biofilm at a rate of 80 % and to varying degrees, 12 isolates are of strong formation of the biofilm, 11 isolates are of medium formation, 13 isolates are weakly formed while 9 isolates were not forming the biofilm\u00a0\u00a0<\/span><\/h3>\n

      The effect of tryptophan amino acid in the form of D and L isomerism was studied using three concentrations (3, 1.5, 0.75) mM on the biofilm formation of three bacterial isolates differing in their biofilm composition (weak, medium, and strong), the results showed that the effect of the amino acid Tryptophan on The biofilm formation was a moderate effect, and the isomeric forms D and L are equally effective in inhibiting the biofilm of P. aeruginosa\u00a0 and\u00a0 when both the D form and the L form were mixed in equal proportions, no inhibitory activity was observed higher than using the isomeric form alone<\/span><\/h3>\n

      A graded effect of acetic acid (2.5 – 0.009) % on biofilm formation was also studied, the acetic acid showed anti-biofilm activity and that the rate of inhibition rate was 65.7 % at concentration 2.5%\u00a0<\/span><\/h3>\n

      As for the molecular study, the two genes ppyR and algD, which play an important\u00a0 role in the formation of the biofilm, were detected by using the PCR in 22 bacterial isolates , The results showed that 100% of the bacterial isolates possessed the ppyR gene with a molecular weight of 160 base pairs, while 95.4% of the isolates possessed the alg D gene and a molecular weight of 1310 base pairs.<\/span><\/h3>\n
